Sensoreal PSP Alert™ Water Test is a rapid immunochromatographic lateral flow assay designed for the qualitative and semi-quantitative screening of saxitoxin and related paralytic shellfish toxins (PSTs) in drinking water samples. The assay uses a competitive immunoassay format. The prepared drinking water sample is incubated with a lyophilized antibody-gold nanoparticle conjugate. If saxitoxin (or a recognized saxitoxin analogue) is present in the sample, it binds to the conjugate antibodies, reducing the amount of free antibody-gold nanoparticle conjugate available to bind at the test line. The mixture is then applied to the lateral flow cassette, where it migrates across a membrane containing an immobilized saxitoxin-protein conjugate at the test line. A control line must appear for the test to be considered valid, confirming proper sample migration and assay performance. Test-line intensity is inversely related to toxin concentration: as the concentration of saxitoxin (or recognized saxitoxin analogues) increases, the intensity of the test line decreases. A negative result is indicated when the test line color is equal to or greater than the intensity of the control line, indicating that the assay did not detect saxitoxin (or a recognized saxitoxin analogue) at or above the method detection limit. Conversely, a test line with color below that of the control line intensity indicates a presumptive positive result. Presumptive positive samples may be confirmed using ELISA, HPLC, or another appropriate confirmatory analytical method.
