Sensoreal DSP Alert™ is a rapid immunochromatographic lateral flow designed for the qualitative and semi-quantitative screening of Okadaic Acid and related diarrhetic shellfish toxins (DSTs) in bivalve mollusks (mussels, oysters, scallops, cockles and razor clams). The assay uses a competitive immunoassay format. Okadaic Acid is extracted from homogenized shellfish tissue using a 100% methanol solution. The extract is then diluted and incubated with a lyophilized antibody–gold nanoparticle conjugate. If Okadaic Acid (or a recognized Okadaic Acid analogue) is present in the sample, it binds to the conjugate antibodies, reducing the amount of free antibody–gold complex available. The mixture is then applied to the lateral flow cassette and migrates across a membrane containing immobilized Okadaic Acid-protein at the test line. A control line must appear for the test to be considered valid, confirming proper sample migration and assay performance. Test-line intensity is inversely proportional to the toxin concentration: as the concentration of Okadaic Acid (or recognized Okadaic Acid analogues) increases, the intensity of the test line decreases. A negative result is indicated when the test line color is equal to or greater than intensity of the control line, indicating that the assay did not detect Okadaic acid (or a recognized Okadaic Acid analogue) at or above the method detection limit. Conversely, a test line with color below that of the control line intensity indicates a presumptive positive result. Presumptive positive samples may be confirmed using ELISA, HPLC, or another appropriate confirmatory analytical method.
